Abstract:
Methods are provided for evaluating estrogenic and antiestrogenic effects of candidate therapeutic agents. Such methods are generally based on assays to detect modulation of estrogen-regulated marker expression in one or more specific cell types. Agents identified using such methods may be used, for example, in the prevention and treatmnt of diseases such as osteoporosis, cardiovascular disease and cancer. In addition, the gene discovery approaches discussed have identified a unique gene profile for estrogen regulation in vascular endothelial cells. This gene profile will allow characterization of the effects of potential SERMs in the cardiovascular system. This gene profile and the assays established with these genes will enable more extensive evaluation of tissue specific properties of SERM compounds and provide a better understanding of cardiovascular effects of SERMs.
